1. Poised enhancers can be subclassified based on presence or absence of H3K27me3 and H3K9me3 from Epigenetic signatures distinguish multiple classes of enhancers

2. H3K4 tri-methylation provides an epigenetic signature of active enhancers:

- H3K4me3-to-H3K4me1 ratio decreases with the enhancer's strength
- gain of H3K4me2/3 -> induction of genes associated w. T-cell development & loss of H3K4me2/3 -> repression of these genes
- concomitant presence of the three H3K4 methylation states (H3K4me1/2/3) was found to clearly reflect the activity of bona fide T-cell gene enhancers
